Zusammenfassung:IntroductionDiabetes mellitus (DM) is deeply associated with the risk of heart failure, especially heart failure with preserved ejection fraction (HFpEF). It has been assumed that HFpEF results from altered diastolic properties. Thus, we examined whether glucose concentration affects the diastolic and systolic force and intracellular Ca (Cai) in normal and DM rat myocardium.MethodsRats were given a subcutaneous injection of 55 mg/kg streptozotocin (Stz-rats) or solvent (Ctr-rats). Trabeculae were dissected from both rat hearts. Force was measured with a strain gauge, Cai with fura-2 and a CCD camera, and sarcomere length with a laser diffraction technique (24°C).ResultsBlood glucose in Stz-rats was higher than that in Ctr-rats (n=9, p
