Zusammenfassung:We predict that SDB in DIO will be treated by leptin-loaded EVs. The effectiveness of EV delivery to the brain in DIO and control lean mice was first assessed. Second, we examined how leptin-loaded EVs affected sleep and breathing in DIO and lean animals in comparison to free leptin and a placebo. The Johns Hopkins University and the University of North Carolina Animal Use and Care Committees authorized all protocols, which complied with the American Physiological Society's Guidelines for Animal Studies. The data supplement includes a detailed description of the methods. In our mouse model of SDB, ventilation is increased by leptin-loaded EVs. Treatment options for SDB and other disorders brought on by defective neural control of breathing include the delivery of leptin and other medicines with limited BBB penetration using EVs produced from macrophages.
