•46,503 home-based SARS-CoV-2 lateral flow tests were performed by hospital staff.•328 (0.7%) of tests were positive.•The positive predictive value was 96% and false positive rate 0.03%.•LFDs were less sensitive than PCR, but detected cases with higher viral loads.
