Zusammenfassung:The authors report here an alternative, sensitive, bioluminescent-enhanced detection method for DNA dot-hybridization. Immobilized DNA is hybridized with a biotinylated probe and incubated with a preformed streptavidin biotinylated alkaline phosphatase complex. Bound complex liberates D-luciferin from the substrate D-luciferin-O-phosphate, and D-luciferin is oxidized by luciferase (Photinus pyralis ) under light emission. Emitted light exposes a sensitive photographic film (Kodac, Tri X-Pan). Thus specifically bound alkaline phosphatase complex can be visualized after development.
