Trypsin variants with redesigned specificity: EK‐trypsin(ogen), produced by replacement of the trypsin “99‐loop” by the KRRK‐sequence from enteropeptidase (enterokinase), exhibits specificity for DDDDK‐containing substrates and is capable of self‐activation.
