Zusammenfassung: | A mathematical pharmacokinetic model, including flow limitations, lipid solubility, protein binding, and metabolism, is used to make a priori predictions of the distribution of thiopental in four body regions. Tissue binding is correlated by means of “effective” protein fractions and bovine serum albumin data. Metabolism is represented by a Michaelis-Menten rate equation. Close agreement with existing experimental data lends confidence in the model as a valuable tool for predictions in a variety of therapeutic situations. |
