Zusammenfassung: | An automated modification of the Bodansky method for alkaline phosphatase and phosphorus in serum is presented. The tests can be performed at a rate of 40 tests/hour. Statistical evaluation showed that the methods are accurate and reproducible.
Highest activity was obtained if a substrate adjusted to pH 9.60 (at 27°) or 9.40 (at 38°) was used. The resultant pH of buffered substrate and serum is between 9.00 and 9.10 (at 27°) and between pH 8.80 and 8.90 (at 38°).
The normal values were found to be 1.3–4.0 units (7.0–21.5 International units) with a mean of 2.5 ± 0.59 S.D.
An alternative procedure is listed which has an increased sensitivity and which eliminates contamination by specimens high in alkaline phosphatase activity but which can be run only at a speed of 20 tests/hour. |
