Zusammenfassung: | Vaccinia virus has been grown in HeLa cell suspension and purified by fluorocarbon treatment. Cellular material was digested by nucleases and by trypsin, and the virus was finally purified by density gradient centrifugation in cesium chloride.
Infected cells have been grown in the presence of uridine-2-C
14 and a coincidence of counts per minute, OD
260, and plaque-forming units has been obtained in the density gradient. Hydrolysis showed that purified virus contained less than 0.2% by weight of RNA as calculated from the presence of uracil-2-C
