Isolation of Rabbit Anti-Rhesus Lymphocyte Antibody Using a Specific Immunoadsorbent

This report describes a rapid, one-step method of purifying both anti-rhesus and anti-human lymphocyte antibodies using a specific immunoadsorbent. The immunoadsorbent was prepared by chemical conjugation of rhesus lymphocyte stroma to Sephadex G-100. Specifically purified antilymphocyte antibody eluted from the immunoadsorbent represented 6% to 9% of the total protein applied to the column. The antibody preparations showed a 10- to 15-fold increase in specific activity (leukoagglutinins/milligram IgG).
