Zusammenfassung: | Hybrid plasmid derivatives of pBR322 that contain yeast (Saccharomyces cerevisiae ) centromere DNA in combination with a yeast DNA replicator (ars1 or ars2) and a suitable genetic marker are stably maintained in yeast through both mitotic and meiotic cell divisions and exhibit typical Mendelian segregation through meiosis. Taking advantage of the unique mitotic stablity of centromere hybrid plasmids (minichromosomes) in yeast and the characteristic instability of other ars plasmids lacking centromere sequences, the authors have developed and present here a procedure that should permit the isolation of centromere DNA sequences of any yeast chromosome directly from a yeast recombinant DNA library. |
