Zusammenfassung: | The adapter subclass of src homology (SH)2 domain-containing proteins consists largely of noncatalytic protein modules and functions downstream of tyrosine kinases, linking receptors or other tyrosine phosphorylated proteins to separate effectors. We have recently cloned a novel SH2 domain-containing adapter, designated Grb14, related in both molecular architecture and sequence homology to Grb7 and Grb10. The structural features that define this emerging family of signaling proteins are an N-terminal region containing a conserved proline-rich peptide sequence, a central region that exhibits significant homology to the Caenorhabditis elegans protein F10E9.6/mig 10 and that also encompasses a pleckstrin homology (PH) domain, and a C-terminal SH2 domain. |
