Zusammenfassung: | A simple, specific and sensitive ion-pair high-performance liquid chromatographic analysis was developed for aminopyrine, sulpyrine and their metabolites (4-methylaminoantipyrine, 4-aminoantipyrine, 4-acetylaminoantipyrine and 4-formylaminoantipyrine) in rabbit plasma. Following deproteinization with acetonitrile and separation on an octadecylsilane-bonded silica column (250×4mm, JASCO Fine SIL C-18), the eluted components were detected with a multiwavelength ultraviolet monitor at 260nm. The mobile phase consisted of 22% (v/v) acetonitrile and 78% (v/v) of an aqueous solution containing 10mM KH2PO4 and 1.24mM tetra-n-butylammonium bromide, adjusted to pH 4.5 with 0.1M phosphoric acid solution. Hexobarbital was employed as an internal standard. Based on 0.1ml of plasma, the detection limits were 0.35μM for 4-acetylaminoantipyrine, 0.35μM for 4-formylaminoantipyrine, 0.7μM for sulpyrine, 1μM for 4-aminoantipyrine, and 2μM for 4-methylaminoantipyrine at the signal-to-noise ratio of 5 : 1. |
