The initiation of TMV infection in isolated protoplasts by polyethylene glycol

Tobacco mosaic virus (TMV) particles bind to isolated tomato protoplasts and following treatment with polyethylene glycol (PEG) initiate infection. Studies indicate that the procedure involves two steps, TMV binding to the plasmalemma, followed by PEG-initiated infection. The percentage of protoplas...

Zusammenfassung:Tobacco mosaic virus (TMV) particles bind to isolated tomato protoplasts and following treatment with polyethylene glycol (PEG) initiate infection. Studies indicate that the procedure involves two steps, TMV binding to the plasmalemma, followed by PEG-initiated infection. The percentage of protoplasts infected is routinely ca. 60% and mean virus production per infected protoplast is ca. 1.5 × 10 6 TMV particles. It is suggested that PEG may induce TMV/plasmalemma fusion.
