Application of models of pharmacological intervention for the optimization of clinical trial designs

Examples are presented that illustrate the use of simulations for clinical trial design optimization. Also discussed are more formal design optimization approaches using Bayesian decision theory, and adaptive trial designs in which PK/PD modeling and Bayesian decision analysis are used for decisions during the trial such as dose assignment and whether to stop the trial.
