Zusammenfassung:A cDNA (cel2) coding for an exo-type cellobiohydrolase was isolated from the basidiomycete Irpex lacteus strain MC-2. The cel2 cDNA was expressed in a heterologous host Aspergillus oryzae by using an expression vector pNAN-8142. A recombinant cellobiohydrolase was secreted in culture fluid of A. oryzae transformant cells with the aid of a signal sequence of the cell gene. The recombinant cellobiohydrolase purified from the culture fluid exhibited similar enzymatic properties to those of a major cellobiohydrolase (Ex-1) previously purified and characterized from Driselase (a commercial enzyme produced by strain MC-2).
