Zusammenfassung:This research was part of research on exploration of microbial chitinolytic enzymes, their application to produce chitooligosaccharides and their bioactivities. The aim of the research was to evaluate the antibacterial of chitooligosaccharides produced from chitosan by using a commercial non-specific cellulase from Trichoderma reesei. This enzyme was firstly determined its optimal temperature and pH as chitosanase, then used for hidrolyzing chitosan, with the enzyme concentration of 10, 13 dan 15 U/g chitosan for I, 2 and 3 hours respectively. The chitooligosaccharide produced was then identified using thin iayer chromatography, and tested its ability in inhibiting the microbial growth. The selected chitooligosaccharide based on its antibacterial activities was further produced in larger scale to determine its viscosity, as well as molecular weight. The result indicated that the cellulase worked optimally as chitosanase at 60 degree C and pH of 6. The chitooligosaccharides produced contained of dimer trimer, tetramer dan pentamer which were able to inhibit the growth of Staphylococcus aureus up to 84%, Salmonella typhosa up to 26%, Bacillus subtilis up to 15% and Escherichia coli up to 5.6%. Hidrolysis of chitosan using 10 U/g chitosan of this enzyme for hour incubation produced oligosaccaharide with viscosity of 2.15 cPs and molecular weight of 323.76 Da.Original Abstract: Penelitian ini merupakan sebagian dari rangkaian penelitian mengenai eksplorasi enzimenzim kitinolitik mikroba dan aplikasinya untuk pembuatan oligomer kitosan (kitooligosakarida) serta uji bioaktivitasnya. Tujuan penelitian ini adalah untuk menguji kemampuan antibakteri kitooligosakarida yang diproduksi dari kitosan dengan menggunakan enzim selulase non spesifik komersial dari Trichoderma reesei. Enzim ini ditentukan terlebih dahulu suhu dan pH optimumnya sebagai kitosanase, kemudian digunakan untuk menghidrolisis kitosan dengan konsentrasi 10, 13, dan 15 U/g kitosan, masing-masing selama 1, 2, dan 3 jam. Kitooligosakarida yang dihasilkan kemudian diidentifikasi dengan menggunakan khromatografi lapis tipis, dan diuji bioaktivitasnya sebagai antibakteri. Kitooligosakarida terpilih berdasarkan aktivitas antibakterinya kemudian diproduksi lagi untuk penentuan viskositas dan bobot molekulnya.
