Zusammenfassung:Objective To investigate the mechanism of Annexin A2 in radioresistant nasopharyngeal carcinoma(NPC) cells by screening for Annexin A2 interacting proteins in radioresistant NPC cells. Methods Screening and identification of Annexin A2 interacting proteins in radioresistant nasopharyngeal carcinoma CNE-2(R743) cells were done by co-Immunoprecipitation(Co-IP) combined with mass spectrometry (MALDI-TOF/TOF). The mass spectra data were searched using NCBInr database with Mascot software. Analysis of the role of Annexin A2 and its interacting proteins in the radiation resistance of nasopharyngeal carcinoma cells by bioinformatics. Results Five proteins were interacting with Annexin A2, which were Calnexin, HSP90, GRP78, Vimentin, HSP27, respectively. Bioinformatics analysis of these proteins and Annexin A2 revealed that GRP78, HSP27 and HSP90 interact with Annexin A2 directly, but Calnexin, Vimentin interact with Annexin A2 indirectly. Conclusion These proteins may interact with Annexin A2 resulting in radiation resistance of nasopharyngeal carcinoma cells.
