Localization of β-glucan synthetase in membranes of Paracoccidioides brasiliensis

To localize beta -1,3-glucan synthetase in membranes of Paracoccidioides brasiliensis, particulate preparations were obtained from both phases of the fungus and separated on sucrose gradients. The yeast glucan synthetase distributed between the 20% and 30% sucrose bands whereas the mycelial enzyme concentrated in the 40% band. In all cases, vanadate-sensitive, azide-insensitive ATPase was found together with glucan synthetase, suggesting that the latter is located in the cytoplasmic membrane.
