Zusammenfassung: | Lipolytic activity of Pseudomonas aeruginosa DRK 9.1 has not been explored yet. This research aims to study the lipolytic activity of this strain. The isolate grown in Bushnell Haas medium with 1 % diesel and 0.5 % glucose or 0.5 % yeast extract were tested. Qualitative lipolytic assay was done on Tween 80 and olive oil agar and quantitative assay was done by titration. Maximum cell number after 48 h was 5.90 x 1010 CFU/mL in diesel and glucose addition medium and 7.47 x 1010CFU/mL in diesel and yeast extract addition. Qualitative lipolytic assay shows fluorescent halos around the colonies. Lipolytic activity after 48 h incubation was 2.71 U/mL in diesel and glucose addition medium and 3.30 U/mL in diesel and yeast extract addition medium. The inoculum possesses lipolytic enzyme but the low lipolytic activity is indicated due to complex diesel components and no emulsification making it difficult to degrade. |
