Zusammenfassung:BackgroundThe human gastrointestinal tract is colonized by billions of commensal microbes - the gut microbiota. Recently, it has become evident that intestinal microbiota plays an essential role in human well-being. Akkermansia muciniphila is a promising bacterium for next-generation probiotics to prevent inflammation. The aim of this work was to evaluate the immunomodulating activity of A.muciniphila.MethodsThis study was conducted using a set of A. muciniphila strains including two strains from healthy baby faeces - A. muciniphila 1061, A. muciniphila 2796 and one strain from the faeces of a healthy adult - A. muciniphila 501. The immunomodulating activity of conditioned medium (CM) from A. muciniphila and on LPS-induced (E. coli O55:B5 (10ng/ml)) IL-8, CXCL10, IL-1b, TLR-4 and TLR-2 mRNA expression was investigated using HT-29 cells model. The cells were grown using a standard protocol and were collected for qRT-PCR after 4 hours of incubation. The first strand of cDNA was synthesized using a reverse transcriptase MMLV according to the instructions of the manufacturer (IDDF2023-ABS-0097 Figure 1).ResultsCM of A. muciniphila 1061 strain increased IL-8 gene expression by 20% (r=0.62, p=0.0002) and CXCL-10 gene expression by 14% (r=0.45, p=0.04) compared with the LPS-induced control (IDDF2023-ABS-0097 Figure 2). CM of A. muciniphila 501 strain increased IL-8 gene expression by 74% (r=0.89, p=0.03) and CXCL-10 gene expression by 68% (r=0.87, p=0.05) compared with the LPS-induced control (IDDF2023-ABS-0097 Figure 3). CM of A. muciniphila 2796 strain decreased IL-8 gene expression by 74% (r=0.74, p=0.04) compared with the LPS-induced control and didn’t change CXCL-10 gene expression significantly (IDDF2023-ABS-0097 Figure 4). CM of all strains A. muciniphila didn’t change IL-1b, TLR-2, TLR-4 gene expression significantly (IDDF2023-ABS-0097 Figure 2-4).
