Zusammenfassung: | To evaluate the genetic variability of 90 lentil genotypes, two different molecular marker techniques namely CAAT Box Derived Polymorphism (CBDP) and Simple Sequence Repeat (SSR) markers were utilized. Average polymorphism information content (PIC) for SSR and CBDP markers were 0.63 and 0.45, respectively. The average genetic distance for CBDP and SSR markers were 0.50 and 0.49, respectively. Among markers tested, the average observed number of alleles (Na) for SSR markers were 4.83 and the average expected heterozygosity (He) for SSR marker was 0.64. The average of effective number of alleles (Ne) for SSR markers 3.01. The average resolving power (Rp) values ranged between 1.51 and 8.67 for SSR and CBDP markers, respectively. Overall, SSR80 showed maximum genetic diversity among the SSRs used. A total of 145 and 135 polymorphic bands were obtained using SSR and CBDP markers, respectively. CBDP and SSR markers showed different efficiency for evaluating DNA polymorphism in lentil. A positive significant correlation (r = 0.89

) for similarity matrix was obtained between CBDP and SSR markers. Neighbor-joining cluster analysis for both markers (CBDP and SSR markers) grouped the genotypes into five distinct clusters. Our results suggested high efficacy for SSR and CBDP markers in fingerprinting of lentil genotypes. This is the first report of using CBDP for genetic diversity analysis in lentil and also a comparison between CBDP and SSR markers among lentil germplasm. Generally, our results are able to show the suitability of CBDP and SSR markers for genetic diversity analysis in lentil for their high rates of polymorphism and their potential for genome diversity and germplasm conservation. |
