EXPRESSION OF TUMOR SUPPRESSOR MIR-142-3P IS REDUCED IN PATIENTS WITH CHOLANGIOCARCINOMA

Cholangiocarcinoma (CCA), bile duct neoplasm, is a rare tumor with a low survival rate. Numerous molecular events contribute to carcinogenesis, including microRNAs, responsible for modulating several genes. In this case, the miR-142-3p, which regulates genes involved in angiogenesis, is highlighted....

Zusammenfassung:Cholangiocarcinoma (CCA), bile duct neoplasm, is a rare tumor with a low survival rate. Numerous molecular events contribute to carcinogenesis, including microRNAs, responsible for modulating several genes. In this case, the miR-142-3p, which regulates genes involved in angiogenesis, is highlighted. –The objective was to evaluate the expression of miR-142-3p in patients with CCA, lifestyle habits and comorbidities, compared to the control group. Eighty-seven individuals were studied, distributed in a study group (SG): 65 patients with OKC (aged between 30 and 87 y, 51% male); Control Group (CG): 22 individuals without liver disease (aged between 24 and 67 y, 82% female). Tissue RNA was extracted, followed by expression analysis by realtime PCR. Lifestyle habits and comorbidities were obtained through an interview and analysis of medical records. An alpha error of 5% was admitted. The miR- 142-3p was reduced in EG? SG? (median=0.4286, minimum=0.002430, maximum=2.934), compared to CG (p=0.0016). There was no association of lifestyle habits and comorbidities with miRNA expression. A higher age group was observed in SG (median age= 62 y), compared to CG (median age=39 y; p= 0.0001). Regarding gender, there was an equivalence in SG (male=51%, female=49%) compared to CG, with female prominence (82%; p=0.0116). Smoking stood out in SG (SG=45%; CG=9%; p=0.0022), which did not occur for alcohol consumption, DM and SAH. The underexpression of miR-142-3p, which acts in the modulation of genes involved in angiogenesis, in patients with CCA, indicates its association with the disease.
