Zusammenfassung:Objectives: Long exposure to alcohol could lead to alcoholic cardiomyopathy and then heart failure. Heart could recover if being absent in asymptomatic stage. However, the processing from asymptomatic stage to symptomatic stage was difficult to be observed. 11C-acetate was a good biomarker to quantitatively calculate myocardial oxygen consumption (MVO2) by Kmono and myocardial blood flow (MBF) by K1. In last study, heavy drinking could induce reduction of myocardial energy supply to demand. The aim of this study was to assess the association of Kmono and K1 between left and right ventricle on the chronic alcoholic patients by 11C-acetate. Methods: Study subjects: In this study, thirty-three subjects with alcohol consumption (48.5 ± 8.9 years) over 10 year's history of alcohol consumption (>2 std/day, 1std = 14g of pure ethanol) and twelve healthy controls (50.4 ± 10.5 years) without drinking history, according to the 2015 to 2020 Dietary Guidelines for Americans. Doppler echocardiography were performed on every subject as screening and the subjects were excluded if had history of hypertension, congenital heart disease and coronary disease. The study protocol was approved by local Institutional Review Board and the written consent form was obtained from each subject. Imaging protocol: PET/CT imaging was performed by a hybrid PET/CT scanner (PoleStar m660, Sinounion, Healthcare Inc., Beijing). Approximately 740 MBq of 11C-acetate was administered intravenously. All subjects underwent a 40-min dynamic scan after the administration of 11C-acetate. A total of 53 frames (15×10, 15×30, 16×60 and 7×120 sec) were corrected for dead time, decay, measured photo attenuation and were reconstructed using 3D OSEM+TOF algorithm on Precision workstation (Sinounion, Healthcare Inc., Beijing), an object space of 192×192×117 and a voxel size of 3.147×3.147×1.8665 mm3. Data analysis: Two radiologists reviewed PET/CT images and drew the long axis from base of left ventricle to calculate polar map, separately. The region of interests on right ventricle were drawn by two radiologists either. The mean of standard uptake value (SUV) were extracted for further calculation. Kmono and K1 were calculated by exponential fitting on two level: global level (left and right ventricle) and 17 segments on left ventricle. Statistical analysis: Results were displayed as mean ± SD.
