Zusammenfassung:[...]it has a saprophytic activity associated with a wide range of hydrolytic and oxidative enzymes that enable this species to grow wherever the suitable source of food and moisture are available (16). Singleplex PCR Master Mix: The Singleplex PCR reactions for detection of 16S rRNA gene of A. niger were performed in 25 pl volumes containing 5.0 pl of nuclease free water, 12pl of GoTaq Green Master Mix 2X containing (GoTaq DNA polymerase supplied in 2X Green GoTaq reaction buffer (pH 8.5), 400 pM dATP, 400 pM dGTP, 400 pM dCTP, 400 pM dTTP, 3 mM MgCh, yellow and blue dyes which function as loading dyes when reaction products are analyzed by agarose gel electrophoresis), 2 pl of 10 pmol F primer and 2pl of 10 pmol R primer and 2 pl of the genomic DNA sample. The present study also agrees with Thammahong et al., 2015 results who found that A. niger was the most species isolated from ear infection followed by A. fumigatus and A. flavus. [...]the results are in agreement with (2) who found that the A. flavus was the most common fungus in otomycosis followed by A. niger, A. fumigatus and A. nidulans. The isolation and identification of A.niger by using conventional methods include direct examination by 10% KOH, culture on SDAC ,macroscopic examination of the colony include (growth rate, color of colony, texture and microscopic examination of the colony include (spoer, chonidiophore, vesicle) which were go togther with study conducted by (1) showed that the fungal culture were postive in 39(78%) patients while11 (32%) revealed negative fungal culture ,the most common type of fungal isolated were A.niger 19(32%) patients was detected by the conventional methodds of direct examination, culture on SDAC, Zapex Dox Agar.
