Zusammenfassung: | Enumeration and immunophenotyping of lung myeloid and lymphoid cell subsets were performed by cell surface and intracellular staining with fluorochrome-conjugated antibodies followed by flow cytometry analysis. Improved fungal clearance in IL-10-/- mice coincided with the following alterations in immune phenotype (relative to infected IL-10+/+ mice): 1) increased lung leukocytes; 2) enhanced classical/DC1 activation of CD103+ cDC, CD11b+ cDC, and monocyte-derived DC (as evidenced by increased expression of CD40, CD80, CD86, iNOS or IL-12 and decreased expression of arginase 1 and CD206); 3) increased numbers of CD4+ and CD8+ T cells; 4) increased Th1 and Th17 polarization (but no change in Th2 or Treg polarization; assessed by percent expression of T-bet+, IFNy+, RORYt+ IL-17A+, GATA3, and FoxP3); and 5) enhanced classical/M1 activation of exudate (but not alveolar) macrophages (based on upregulated expression of CD80, CD86 and iNOS and downregulated expression of CD206). |
