인체 각막표층의 MUC1 의 생성 및 분포

각막 표면에 존재하는 뮤신중 MUC1은 결막 및 각막 상피세포가 생산하여 세포막 점액을 구성케 하는 것으로 알려져 있다. 저자들은 이 뮤신 MUC1의 인체 각막에서의 생성양상 및 존재 양상을 알아보고자 하였다. 인체 적출 안구의 표면에 4% paraformaldehyde가 포함된 고정액을 점안한 후 각막만을 분리하여 sucrose 처리하고,이를 급속 냉동한 후 연속절편으로 만들어 면역조직화학 염색, 면역 형광 염색, in situ hybridization으로 관찰하였다. 면역조직화학 염색상, 인체 각막의 표면 상피세포막에 MUC1이 관찰되었다. 면역 형광 염색상, 이 표면 상피세포막의 MUC1은 세포마다 다른 양의 분포를 보였다. In situ hybridization상 MUC1의mRNA는 각막의 상피세포 전층에서 발견되었다. 뮤신 MUC1은 인체 각막 뮤신층의 일부로서의 표층으로 이동한 표면 상피세포의 첨부 세포막에 존재하며, 각 세포마다 그 양에는 차이를 보였다. 이는 MUC1의 발현이 상피세포의 수직이동과 관련되며, 각각의 세포에 따라 영향을 받는 것으로 보인다(한안지 42:145~151, 2001). Corneal and conjunctival squamous epithelial cells have been known to express the mucin MUC1. We attempted to reveal the expression and localizational characteristics of the membrane-spanning mucin MUC1 as a component of the mucous layer in the human corneal epithelium. An antibody to the MUC1 was used to detect the MUC1 on the corneal epithelium by immunohistochemistry and immunofluorescent staining. In situ hybridization was performed to determine the distribution of MUC1 mRNA in the ocular surface. Immunohistochemically, the MUC1 mucin was observed along the apical membranes of the corneal epithelium. According to immunofluorescent staining, cells varied in the amount of mucin MUC1. Expression of MUC1 mRNA was observed in all layers of the corneal epithelium. The MUC1 mucin synthesized by the corneal epithelia exists on the apical membrane of the superficial cells. The amount of MUC1 may vary with the vertical migration and the activity of the cells(J Korean Ophthalmol Soc 42:145~151, 2001).
