Zusammenfassung:Diazepam, a mejor anxiolytic, administration exhibited learning and memory impairment in rodents. The aim of this study is to evaluate the effects of chronic diazepam administration and its withdrawal on spatial learning in rats, using eight arms radial-maze. Each rat was trained one trial per day for 35 days. Diazepam (0, 1.0, 2.0mg/kg s.c.) was administered 30 min before each trial during the 1-28 trials. In the 29-35 trials, diazepam administration was not carried out to evaluate diazepam withdrawal. As the results, chronic diazepam administration impaired spatial learning, because reference error was increased diazepam groups compare with control group. But diazepam withdrawal caused to reverse the impairment of spatial learning. Win-Shift behavior was caused by chronic diazepam administration and disappeared notably by diazepam withdrawal. These results suggest that chronic diazepam administration influenced to formation of spatial learning, and diazepam withdrawal reversed chronic diazepam administration induced impairment of spatial learning in rats.
