Zusammenfassung:We found that Hyaluronic acid poly sulfated ester (HAPS) is a potent kallikrein-kinin inhibitor. Recently, many reports indicated bradykinin produced by the kallikrein-kinin system is an important mediator of allergic rhinitis. We investigated the effect of HAPS on experimental allergic rhinitis in guinea pigs. Guinea pigs were actively sensitized by inhalation of 1% ovalbumin for 10 min on two occasions, separated by one week. Various immediate responses induced by administration of ovalbumin into the nasal cavity were investigated as follows: Changes in intranasal pressure as nasal obstruction, exudation of dye into nasal lavage fluid as pituita and, frequency of sneezes as sneezing were determined. In the sensitized guinea pigs, intranasal pressure and exudation of dye into the nasal lavage fluid were increased and onset of sneezes were observed after antigen challenge. Bilateral intranasal administration of HAPS at 20μg inhibited the increase in intranasal pressure and exudation of dye as well as sneezes. Ketotifen at 20μg inhibited the exudation of dye and sneezes but not the intranasal pressure. The above results indicated that HAPS inhibited immediate increase in intranasal pressure and exudation of dye as well as sneeze in experimental allergic rhinitis of guinea pigs, suggesting the clinical efficacy of HAPS against nasal obstruction, pituita and sneeze in the treatment of allergic rhinitis.
