Zusammenfassung:Tobacco streak virus (TSV), the causal agent of cotton necrosis, is an emerging disease of national significance in India. Owing to its significance, cotton plants cultivated in Tamil Nadu, India were surveyed for cotton necrosis during 2015-2016. The samples collected from different cotton-growing tracts of Tamil Nadu were observed for the presence of symptoms associated with TSV infection in cotton (Gossypium hirsutum and Gossypium barbadens). Serological assay by direct antigen coating enzyme-linked immunosorbent assay (DAC-ELISA) revealed the presence of TSV in leaf, square, stem, root, petiole and pollen grains of both asymptomatic and symptomatic samples of variety CO 14 of cotton. Similarly, the presence of TSV in different plant parts including root, stem, leaf, pollen grains and squares in both asymptomatic and diseased plants was confirmed through reverse transcriptase polymerase chain reaction (RT-PCR). All the samples collected from both asymptomatic and symptomatic samples yielded the expected PCR product of 929 bp including the coat protein gene of TSV and thus confirmed the systemic nature of the virus. Since parthenium (Parthenium hysterophorus) serves a a reservoir of TSV, attempts were also made to confirm the systemic nature of TSV in asymptomatic parthenium plants. DAC-ELISA and RT-PCR based detection tools confirmed the presence of TSV in the asymptomatic leaf, stem and pollen grains.
