Positive amplification in reverse transcription polymerase chain reaction (RT-PCR), with two sets of degenerate primers, was used to detect viruses in the families Flexiviridae and Closteroviridae. Sequence analyses of the RT-PCR products identified Cherry virus A (CVA), Cherry necrotic rusty mottle virus (CNRMV), Cherry green ring mottle virus (CGR-MV), Little cherry virus 1 (LChV-1) and Plum bark necrosis and stem pitting associated virus (PBNSPaV). [Nel corso del 2003, in indagini su frutteti della California coltivati a ciliegio dolce [Prunus avium (L.) L. cv Bing], sono state raccolte foglie da piante che presentavano sintomi di necrosi delle nervature, maculature necrotiche rugginose, foglia necrotizzata o frutto punteggiato. Sono stati pure prelevati campioni da piante con lesioni necrotiche a carico delle foglie (P. serrulata Lindl. Cv Kwanzan) e macchie fogliari clorotiche (P. avium x P. pseudocerasus cv Colt) da blocchi di Prunus per la catalogazione delle malattie. Tutti i campioni provenienti dalle piante malate, ma non da quelle sane, producevano bande di RNA a doppia elica ad alto peso molecolare a seguito di estrazione dai tessuti, purificazione ed elettroforesi in gel di poliacrilamide. E´ stata utilizzata l´amplificazione positiva nella RT-PCR, con due gruppi di primer degenerati, per diagnosticare i virus delle famiglie Flexiviridae e Closteroviridae.
