Zusammenfassung:Leaves were collected during 2003 from trees displaying symptoms of vein necrosis, necrotic rusty mottle, necrotic leaf or marked fruit in surveys of sweet cherry [Prunus avium (L.) L. cv Bing] orchards in California. Samples were also taken from trees with leaf necrotic lesion (P. serrulata Lindl. cv Kwanzan) and leaf chlorotic spots (P avium x P pseudocerasus cv Colt) from virus disease indexing blocks of Prunus. All of the diseased, but none of the healthy, collections yielded high molecular weight double stranded RNA bands following tissue extraction, purification and electrophoresis in acrylamide gel. Positive amplification in reverse transcription polymerase chain reaction (RT-PCR), with two sets of degenerate primers, was used to detect viruses in the families Flexiviridae and Closteroviridae. Sequence analyses of the RT-PCR products identified Cherry virus A (CVA), Cherry necrotic rusty mottle virus (CNRMV), Cherry green ring mottle virus (CGR-MV), Little cherry virus 1 (LChV-1) and Plum bark necrosis and stem pitting associated virus (PBNSPaV). This is the first report of the occurrence of the viruses CVA, CNRMV and LChV-1 in California [Nel corso del 2003, in indagini su frutteti della California coltivati a ciliegio dolce [Prunus avium (L.) L. cv Bing], sono state raccolte foglie da piante che presentavano sintomi di necrosi delle nervature, maculature necrotiche rugginose, foglia necrotizzata o frutto punteggiato. Sono stati pure prelevati campioni da piante con lesioni necrotiche a carico delle foglie (P. serrulata Lindl. Cv Kwanzan) e macchie fogliari clorotiche (P. avium x P. pseudocerasus cv Colt) da blocchi di Prunus per la catalogazione delle malattie. Tutti i campioni provenienti dalle piante malate, ma non da quelle sane, producevano bande di RNA a doppia elica ad alto peso molecolare a seguito di estrazione dai tessuti, purificazione ed elettroforesi in gel di poliacrilamide. E´ stata utilizzata l´amplificazione positiva nella RT-PCR, con due gruppi di primer degenerati, per diagnosticare i virus delle famiglie Flexiviridae e Closteroviridae. L´analisi delle sequenze dei prodotti RT-PCR ha portato a identificare il Virus A del ciliegio (CVA), il Virus della maculatura necrotica rugginosa del ciliegio (CNRMV), il Virus della maculatura anulare verde del ciliegio (CGR-MV), il Virus del ciliegio piccolo 1 (LChV-1) e il Virus associato alla necrosi della corteccia e alle maculature del fusto del susino (PBNSPaV).
