Zusammenfassung: | The study was designed to investigate the effect of RO 44-9883 on bleeding time when administered during a thoracotomy, on platelet function, and on cyclic flow variations following coronary occlusion and reperfusion (REP). Fourteen animals had thoracotomy, catheterinduced intimal injury to the LAD and 55 mm LAD occlusion followed by 5 h of REP. Blood flow was monitored at the occlusion site with an electromagnetic flowmeter. RO 44-9883. 250 μg/kg bolus followed by 3 μg/kg/min infusion or placebo was administered, at random, 5 min before REP for 5 h. Results Basal bleeding time was the same in treated animals and in controls (100±10 vs 88±13 sec), but treated animals showed a significant increase in bleeding time at 15 min (232±41 vs 105±22), 1 hour (285±77 vs 86±14) and 2 hours of REP (198±57 vs 81±7).
