The optimized method for extracting RNA of milk fat includes the following steps: S1. sample acquisition; S2. homogenization; S3. upper lipid layer removal; S4. incubation; S5. isolation; S6. uniform mixing; S7. re-centrifugation; S8. precipitate washing; S9. centrifugation for purification; S10. air-drying and dissolving of RNA; and S11. final assay. After the pyrolysis by a Trizol reagent, the milk fat is re-centrifuged to ensure the purify of the RNA extracted. In the process of precipitation of RNA by an isopropyl alcohol solution, RNase-free Glycogen reagent is added to improve the concentration, purify, and integrity of RNA. |
