Zusammenfassung:	Provided is a blocking sequence, a kit thereof, and a method for using same. The blocking sequence comprises a blocking sequence 1, a blocking sequence 2, a blocking sequence 3, and a blocking sequence 4. The blocking sequence 1 is complementary to a sequencing joint P5 and used for blocking the sequencing joint P5 at the 5' terminal of a first chain when a capture library is prepared in a target area. The blocking sequence 2 is complementary to an Rsp1 sequence and used for blocking the Rsp1 sequence in a 5'-to-3' direction on the first chain when the capture library is prepared in the target area.
