According to the present invention, a key enzyme mutant gene with a high efficiency is screened out from an iterative key enzyme gene mutation library by means of weakening the expression of the key enzyme gene of the high value-added metabolite, using the automation technology, and in combination with a mutation-screening-mutation closed-loop protein evolution method.
