AUTOANTIGENIC PEPTIDES (CALVICIFIV), PRESENTED BY TOLEROGENIC DENTRITIC CELLS, USEFUL FOR THE PERSONALIZED TREATMENT OF RHEUMATOID ARTHRITIS

The present invention provides an immunomodulatory composition useful for the treatment or prevention of joint damage comprising at least a set of peptides possessing an amino acid sequence having at least 80%, 85% and 90% sequence identity with the peptides corresponding to SEQ ID No.1, SEQ ID No 2, SEQ ID No.3 and SEQ ID No.4 and a method for the treatment or prevention of joint damage comprising the steps of a) extract monocytes from a patient with a rheumatological disease; b) culture the monocytes extracted in the previous step in AIM-V medium with GM-CSF and IL-4; c) wash the monocytes and add dexamethasone; d) load the tDCs with the immunomodulatory composition comprising autoantigenic peptides; e) add MPLA; and f) incorporate the tDCs loaded with autoantigenic peptides into the patient. The present invention includes methods for the treatment or prevention of rheumatological diseasecomprising a wide range of tDCs performed by different protocols. La présente invention concerne une composition immunomodulatrice utile pour le traitement ou la prévention des lésions articulaires comprenant au moins un ensemble de peptides possédant une séquence d'acides aminés ayant au moins 80 %, 85 % et 90 % d'identité de séquence vis-à-vis des peptides correspondant à SEQ ID no : 1, SEQ ID no : 2, SEQ ID no : 3 et SEQ ID no : 4 et un procédé de traitement ou de prévention des lésions articulaires comprenant les étapes de a) extraction des monocytes d'un patient présentant une maladie rhumatologique ; b) culture des monocytes extraits à l'étape précédente dans du milieu AIM-V avec GM-CSF et IL-4 ; c) lavage des monocytes et ajout de dexaméthasone ; d) charge des tDC avec la composition immunomodulatrice comprenant les peptides autoantigéniques ; e) ajout de MPLA ; et f) incorporation des tDC chargés des peptides autoantigéniques dans le patient. La présente invention comprend des procédés de traitement ou de prévention de la maladie rhumatologique comprenant une large plage de tDC effectués par différents protocoles.
