The present invention provides a method for producing a recombinant CRM197 protein soluble in the cytoplasm, the method comprising a step of culturing recombinant E. coli anchoring an expression plasmid carrying a gene sequence coding for a recombinant CRM197 protein under a condition suitable for expression of the recombinant CRM197 protein which has a histidine-tag attached to the C-terminus of CRM197 and a maltose-binding protein (MBP) attached to the N-terminus of CRM197. L'invention concerne une méthode de production d'une protéine CRM197 soluble à haut rendement par expression excessive de la protéine dans le cytoplasme d'E. coli. La présente invention concerne une méthode de production d'une protéine CRM197 recombinante soluble dans le cytoplasme, ladite méthode comprenant une étape de culture d'E. coli recombinant par ancrage d'un plasmide d'expression portant une séquence génique codant pour une protéine CRM197 recombinante dans des conditions appropriées pour l'expression de la protéine CRM197 recombinante qui a une étiquette histidine attachée à l'extrémité C-terminale de CRM197 et une protéine de liaison au maltose (MBP) fixée à l'extrémité N-terminale de CRM197. 대장균(E. coli)으로부터 세포질내에서 용해성의 CRM197 단백질을 과량 발현시켜 고수율로 수득하기 위한 생산 방법이 개시된다. 본 발명은 재조합 CRM197 단백질 발현에 적합한 조건에서, CRM197의 C-말단에 히스티딘-태그(histidine-tag)가 부가되고 N-말단에 말토오스-결합 단백질(Maltose-binding protein; MBP)이 부가된 재조합 CRM197 단백질을 인코딩하는 유전자 서열을 포함하는 발현 플라스미드를 포함하는 재조합 대장균을 배양하는 단계를 포함하여, 세포질내에서 용해성인 재조합 CRM197 단백질을 생산하는 방법을 제공한다.
