The present invention relates to atovaquone prodrug compound of formula (I). Accordingly, present invention provides a process involving condensation of Atovaquone (II) with 5-methyl-4- chloromethyl dioxalone (III) in suitable solvent system and optionally followed by distillation and crystallization to provide Atovaquone prodrug compound of formula (I) in high yields, purity, and suitable for large-scale manufacture. La présente invention concerne un composé promédicament d'atovaquone de formule (I). Elle concerne un procédé qui consiste à condenser de l'atovaquone (II) avec de la 5-méthyl-4-chlorométhyl dioxalone (III) dans un système de solvant approprié, puis éventuellement à effectuer une distillation et une cristallisation afin d'obtenir un composé promédicament d'atovaquone de formule (I) à haut rendement, très pur, adapté à une production à grande échelle.
