Zusammenfassung: | The present invention relates to the use of monoolein-based cationic lipid systems for the complexation, transport and transfection of genetic material into target cells. The lipid systems are prepared by conjugating a neutral lipid, monooleoyl-rac glycerol (monoolein), with cationic surfactants derived from the dioctadecyldimethylammonium (DODAX) group, such as dioctadecyldimethylammonium bromide (DODAB), dioctadecyldimethylammonium chloride (DODAC) and dioctadecyldimethylammonium phosphate (DODAP). This lipofection method exhibits high levels of biocompatibility and reduced acquisition costs, and can be used for molecular biology research purposes (in vitro), as well as for gene therapy for medical purposes (in vivo).
La présente invention concerne l'utilisation de systèmes lipidiques cationiques à base de monooléine dans la complexation, le transport et la transfection de matériel génétique dans des cellules cibles. Les systèmes lipidiques sont préparés par conjugaison d'un lipide neutre, le monooléoyl-rac-glycérol (monooléine), avec des tensioactifs cationiques de la famille du dioctadécyldiméthylammonium (DODAX) (bromure de dioctadécyldiméthylammonium (DODAB), chlorure de dioctadécyldiméthylammonium (DODAC) et phosphate de dioctadécyldiméthylammonium (DODAP)). Ce procédé de lipofection trouve une application dans le domaine de la recherche, en biologie moléculaire (application in vitro), ainsi que dans celui de la médecine, en thérapie génique (application in vivo), et permet d'obtenir des niveaux élevés de biocompatibilité ainsi que de faibles coûts d'acquisition.
A presente invenção diz respeito à aplicação de sistemas lipídicos catiónicos à base de Monooleína, na complexação, transporte e transfecção de material genético em células alvo. Os sistemas lipídicos são preparados através da conjugação do lípido neutral monooleoil-rac-glicerol (Monooleína) com os tensioactivos catiónicos da família de Dioctadecildimetilamónio (DODAX) (Brometo de Dioctadecildimetilamónio (DODAB), Cloreto de Dioctadecildimetilamónio (DODAC) e Fosfato de Dioctadecildimetilamónio (DODAP)). Este método de lipofecção é aplicável tanto no domínio da investigação em Biologia Molecular (aplicação in vitro), como na área da saúde em Terapia Génica (aplicação in vivo), apresentando elevados níveis de biocompatibilidade e reduzidos custos de aquisição. |
