Zusammenfassung: | The present invention provides a method for producing human chimeric antigen receptor T-cell enriched of stem cell-like memory T-cells, comprising: (1) Providing human peripheral blood mononuclear cells (PBMC); (2) Human T cell isolation: adding magnetic beads coated with anti-CD3/CD28 antibodies to human PBMC to obtain human T cells/ magnetic beads complexes; (3) Human T cell activation: culturing the human T cells/ magnetic bead complex to obtain activated human T cells/ magnetic beads complexes; (4) Lentivirus transfection: adding lentivirus to the activated human T cells/ magnetic beads complexes to obtain human CAR-T cells/ magnetic beads complexes; (5) Magnetic beads removal: removing the magnetic beads from human CAR-T cells/ magnetic beads complexes after lentivirus transfection to obtain human CAR-T cells; and (6) human CAR-T cells expansion: further culturing the human CAR-T cells to obtain the human CAR-T cells enriched of stem cell-like memory T-cells.
