Method for the preparation of 1-(3,3-diphenyl-3-cyanopropyl)-4- (1-piperidyll)piperidine-4-carboxamide

Process for the preparation and isolation of 1-(3,3-diphenyl-3-cyanopropyl)-4-(1-piperidinyl)piperidine-4- carboxamide (piritramide) by the reaction of 4-bromo-2,2- diphenylbutyronitrile with 4-(1-piperidinyl)piperidine-4- carboxamide in the presence of a base and a solvent selected from N,N-dimethylformamide or aliphatic alcohol with the boiling point in the range of 100 to 140 øC. Subsequently, piritramide is isolated from the reaction mixture by means of an addition salt thereof which is formed by the addition of an organic or inorganic acid. The piritramide addition salt is converted into piritramide by using inorganic base.
