Zusammenfassung: | The vector being inserted into the cell closed to the suitable bacterial host containing repressor C1 with a low thermal stability, causes an increase of the temperature related to the host cell to 42 Celsius degrees, in the case this repressor is being deactivated, the gene expression of the inserted vector and the production of the polypeptide coded by the gene containing the DNA molecule in a form of two fibres, containing the following components in the direction 5'->3': promoter and operator PLOL closed to bacteriophage lambda, instead of using N for establishment of N antiterminator protein generated by the host cell, the DNA sequence containing the bond place for ribosome used to establish nRNA to ribosome closed to the host cell, where instead of the ribosome bond, there is (i) mutation ribosome bond place CII based on the bacteriophage lambda with the sequence TAAGGAAGTACTTACAT; ATTCCTTCATGAATGTA or (ii) synthetic oligo-nucleotide with the sequence TAAGGAAGTACTTACAT; ATTCCTTCATGAATGTA or (iii) of the reweighing main protein gene closed to bacteriophage lambda having the sequence TTTTTTTACGGGATTTTTTTATG; AAAAAAATGCCCTAAAAAAATAC, initiator codon and the area closed to the restrictive enzyme used for inserting of the required gene into the vector in the phase with the initiator condon ATG, it also includes the sequence DNA containing replication origin related to the bacterial plasmid pBR322 being able of autonomous replication in the host cell and the DNA sequence containing gene, connected with a phenotypic feature being able of the selection identification, which is manifested, in the case the vector is present in the host cell. |
