The invention comprises the 3-methyl, 3-ethyl and 3-n-propyl ethers of 17α-(1-propynyl)- estradiol, and the preparation thereof (a) from the corresponding 17-ones by reacting with Li 2 C = C = CLi 2 ; or (b) from the corresponding 17-ones by reaction with CH 3 C#CMgX (X is Br, Cl.or I); or (c) from the corresponding 17α- ethynyl compounds by protection of the 17#-OH group, methylation of the 17α-ethynyl group and liberation of the 17#-OH group; or (d) from the corresponding 17α-propargyl compounds by protection of the 17#-OH group, base isomerization of the 17α-propargyl group and liberation of the 17#-OH group; or (e) from the corresponding 3-ols by alkylation. The compound 3 - (n - butoxy) - 17α - (1 - propynyl) - estra - 1,3,5(10) - trien - 17# - ol is also disclosed. 17α Ethynyl - 17# - (tetrahydropyran - 2- yloxy) - 3 - methoxy - estra - 1,3,5(10) - triene and 17α - propargyl - 17# - (tetrahydropyran-2-yloxy)- 3 - methoxy - estra -1,3,5(10) - triene are prepared from the corresponding 17-ols, and each is converted into 17α - (1 - propynyl) - 17# - (tetrahydropyran - 2 - yloxy) - 3 - methoxy - estra- 1,3,5(10)-triene. The compounds of the invention are said to possess deciduogenic, estrogenic and progestational activities, and may be made up with carriers into pharmaceutical compositions for oral or parenteral administration.
