Invention involves preparing precursor of recombinant insulin by suspending cells E. coli expressing fused proinsulin proteins as inclusion bodies, suspending inclusion bodies in solution containing urea or guanidine HCl, sulfonation of proinsulin fused proteins with sodium sulfonate and sodium tetrathionate, centrifugation of proinsulin S-sulfonate fused protein, its purification by anion-exchange chromatography, dilution in buffer solution and blowing with gaseous nitrogen, mixing with buffer solution containing 2-mercaptoethanol and stirring to obtain reactive refolding-mixture, adsorption purification and elution of human proinsulin with reconstructed chain.
