Zusammenfassung: | The invention relates to a process for preparing Au nanoparticles (AuNP) functionalized with polyhydrazides, meant to be used in biosensor development. According to the invention, the process comprises the following steps: dissolving the polyhydrazide in organic solvent, at concentrations of 0.1...50 mM, dissolving chloroauric acid in water, at concentrations of 0.1...15.0 mM, while maintaining a hydrazide : gold ratio of 3.3:1, gradually adding the chloroauric acid solution in the polymer solution, while stirring, for 5...60 min, collecting the polyhydrazide-grafted AuNP by centrifugation, purification and immobilization of biomolecules on polyhydrazide-grafted AuNP in solution and application/dispersion on surfaces, by means of activation agents, to result in bioconjugates as polymeric matrices with preserved structure and long-term activity suitable for their use as biosensors.
Invenţia se referă la un procedeu de obţinere a unor nanoparticule de Au (AuNP) funcţionalizate cu polihidrazide utilizate în dezvoltarea de biosenzori. Procedeul, conform invenţiei, constă în etapele: dizolvare polihidrazidă în solvent organic, la concentraţii de 0,1...50 mM, dizovare acid colroauric în apă la concentraţii de 0,1...15,0 mM, cu menţinere unui raport hidrazide/aur de 3,3/1, adăugarea treptată a soluţiei de acid cloroauric în soluţia de polimer sub amestecare, timp de 5...60 min, colectare AuNP grefate cu polihidrazide prin centrifugare, purificare şi imobilizarea biomoleculelor pe AuNP grefate cu polihidrazide în soluţie şi aplicare/dispersare pe suprafeţe, prin intermediul unor agenţi de activare, rezultând bioconjugate sub formă de matrici polimerice cu structură conservată şi activitate pe termen lung adecvate pentru utilizare ca biosenzori. |
