Zusammenfassung: | The invention relates to a process for preparing a nanoparticle-based product with oxidative stress reduction properties as well as for preventing the formation of cerebral extracellular β-amyloid plaques characteristic to certain neurodegenerative diseases. According to the invention, the process consists of the following steps: encapsulation of the bioactive ingredient - curcumin - within mesoporous silica particles synthesized in situ by a sol-gel process in aqueous medium, at a temperature of 40°C, in the presence of polyoxyethylene sorbitan monooleate as a surface-active agent, using triethoxy(phenyl)silane and (3-aminopropyl)triethoxysilane as silica sources, addition of Ag nanoparticles (NP) in the sol-gel system with absorption thereof on the surface of the silica particles charged with curcumin, grafting of PEG 750 chains at the surface of the silica particles and maintaining the reaction mixture under stirring for 12 h, at 40°C, to result in functionalized PEG-silica particles charged with curcumin and Ag NP which simultaneously act as active ingredient transport vectors and as β-amyloid filbrillation inhibitors.
Invenţia se referă la un procedeu de obţinere a unui produs pe bază de nanoparticule cu proprietăţi de reducere a stresului oxidativ şi de prevenire a formării plăcilor β-amiloidice extracelulare cerebrale caracteristice unor afecţiuni neurodegenerative. Procedeul, conform invenţiei, constă în etapele de: încapsulare a ingredientului bioactiv curcumină în particule de silice mezoporoasă sintetizate in situβ- printr-un procedeu sol-gel în mediu apos, la temperatura de 40°C, în prezenţă de polioxietilensorbitan monooleat ca surfactant, utilizând ca sursă de silice trietoxi(fenil) silan şi (3-aminopropil)trietoxisilan, adăugare de nanoparticule (NP) de Ag în sistemul sol-gel cu absorbţia acestora pe suprafaţa particulelor de silice încărcate cu curcumină, grefarea lanţurilor de PEG 750 la suprafaţa particulelor de silice şi menţinerea amestecului de reacţie sub agitare, la 40°C, timp de 12 h, rezultând particule funcţionalizate de PEG-silice încărcate cu curcumină şi NP de Ag care acţionează simultan ca vectori de transport pentru ingredientul activ, precum şi ca inhibitori al fibrilării β-amiloidice. |
