Zusammenfassung: | The invention relates to a method of increasing survival and the functionality of implants by stimulating local vascularization in poorly vascularized (subcutaneous) situses with applicability in regenerative medicine for patients with diabetes. According to the invention, the method consists of: producing adult liver cells, infecting them with pancreatic transcription factors in order to induce transdifferentiation of the infected cells, co-implanting the cells in vivo at the subcutaneous level, in an immunodeficient murine medium, with VEGFA co-producing and secreting cells. The VEGFA-producing cells are generated in vitro 2 days prior to in-vivo implantation. Local production and secretion of VEGFA induces vascularization at the implantation site.
Invenţia se referă la o metodă de creştere a supravieţuirii şi funcţionalităţii implanturilor prin stimularea vascularizaţiei locale în situsuri cu vascularizaţie slabă (subcutanat) cu aplicabilitate în medicina regenerativă pentru pacienţii cu diabet. Metoda, conform invenţiei, include: obţinerea celulelor hepatice adulte, infectarea acestora cu factori de transcripţie pancreatici în vederea inducerii transdiferenţierii celulelor infectate, co-implantarea celulelor in vivo la nivel subcutanat, într-un mediu murin imunodeficient, cu celule co-producătoare şi secretoare de VEGFA. Celulele producătoare de VEGFA sunt generate in vitro 2 zile anterior implantării in vivo. Producţia şi secreţia locală de VEGFA determină inducerea vascularizaţiei la locul implantării. |
