Zusammenfassung: | The invention relates to a method for detecting and characterizing the enzymatic activity of the 20S proteasome used for the early detection of medical abnormalities, especially of cancerous or neurodegenerative nature. According to the invention, the method consists in using an electrochemical biosensor with 20S proteasome made by immobilizing, in a single step, the 20S proteasome on electrodes previously modified with antibodies specific to the β 5i subunit and the enzyme complex by crosslinking with glutar aldehyde in the presence of bovine serum albumin, and a peptide substrate marked with 4-amino-7-methyl-coumarin, for detection, by fixed potential chronoamperometry, which enables detection of very small amounts of analyte, with a detection limit of nanomolar range.
Invenţia se referă la o metodă de detectare şi caracterizare a activităţii enzimatice a proteazomului 20S utilizată pentru detecţia precoce a unor anomalii medicale, în special de natură canceroasă sau neurodegenerativă. Metoda, conform invenţiei, constă în utilizarea unui biosenzor electrochimic cu proteazom 20S construit prin imobilizarea într-un singur pas a proteazomului 20S pe electrozi modificaţi în prealabil cu anticorpi specifici subunităţii β 5i a complexului enzimatic prin reticulare cu aldehidă glutarică în prezenţă de albumină serică bovină, şi a unui substrat peptidic marcat pentru detecţie cu 4-amino-7-metil-cumarină, prin cronoamperometrie la potenţial fix, care permite detecţia unor cantităţi foarte mici de analit, cu o limită de detecţie de ordinul nanomolar. |
