The centrifuged ppte is washed two hours with soln. of (NH4)2SO4 (25%, 25g per 100 ml of water), and buffered with Na3PO4/H3PO4, 0.03 M, pH = 5, containing 0.3 M of NaCl and 0.02 per cent of NaN3. Chromatography, and elution, with buffer K2HPO4, 0.02 M, pH = 8, and further chromatography, is followed by elution with 0.1 M NaCl in K2HPO4, 0.02 M, pH = 8. |
