Drug delivery composition

Disclosed is a multiparticulate composition comprising a plurality of beads, each bead comprising: an inert substrate or core; a solubilizing agent in the form of a surface active agent or a pH modulator disposed about the inert substrate or between the semi-permeable layer and the medicament layer, a semipermeable coating; particles of a medicament said particles having an effective average particle size of less than or about 2 μm and a surface stabilizer adsorbed on the surface of the medicament particles. Further disclosed are the methods for making composition in the form of a multiparticulate dosage form, comprising: contacting particles of medicament with at least one surface stabilizer for a time and under conditions sufficient to provide medicament particles having an effective average particle size of less than or about 2 μm; and forming a plurality of beads, each bead comprising: an inert core; a layer of solubilizing agent; a layer of the medicament particles; and a semipermeable coating as the outermost layer.
